Malaria is a parasitic disease caused by Plasmodium, a single-celled eukaryotic organism. The primary species that infect humans include Plasmodium falciparum, Plasmodium vivax, and Plasmodium ovale. Transmission occurs through mosquito vectors and blood, endangering human health. Among these parasites, P. falciparum is the most deadly. The incubation period varies by species, with typical durations of 12–30 days, though it can extend to approximately one year. Onset of the illness brings chills, fever, and sweating; anemia and splenomegaly may also develop. Severe cases may present with coma, profound anemia, and acute renal failure, potentially leading to death. Malaria is found worldwide, predominantly in tropical and subtropical regions such as Africa, Central America, and South America.
Existing detection techniques include blood smear microscopy, antigen testing, and nucleic acid analysis. Recent advances in isothermal amplification enable rapid, simple detection of Plasmodium nucleic acids, making this method ideal for large-scale screening in malaria-endemic areas.
