Epidemiology
Hand, foot, and mouth disease (HFMD) is triggered by enteroviruses, of which 108 serotypes have been identified and classified into groups A through D. The primary causative agents are enterovirus 71 (EV71) and coxsackievirus A16 (CoxA16). The illness predominantly affects children younger than 5 years, presenting with vesicular lesions on the hands, feet, and oral mucosa. A minority of cases may progress to severe complications such as myocarditis, pulmonary edema, and aseptic meningoencephalitis.
Work Flow
Option 1.
For on-site sample collection, use a Sample Release Reagent following the provided directions. Collect oropharyngeal swabs or herpes fluid specimens directly into the reagent. Vortex the mixture thoroughly, incubate at room temperature for 5 minutes, and then invert to mix well. This process extracts the RNA from each sample.
Option 2.
Use a General DNA/RNA Kit (suitable for use with an automatic nucleic acid extractor) and perform extraction as instructed. Use 200 µL of sample and elute in a recommended volume of 80 µL.
Option 3.
Employ a commercial viral RNA mini kit or a virus DNA/RNA extraction kit (catalog no. YDP315-R), adhering strictly to the kit's protocol. Use a sample volume of 140 µL and elute in 60 µL.
