Epidemiology
Over 30 distinct BRAF mutations have been identified, with approximately 90% occurring in exon 15. The most prevalent is the V600E mutation, where a thymine (T) to adenine (A) change at nucleotide 1799 leads to a valine (V) to glutamic acid (E) substitution at codon 600. These mutations are frequently observed in malignancies such as melanoma, colorectal carcinoma, thyroid cancer, and lung cancer. In the context of targeted therapy, assessing BRAF mutation status is essential for identifying patients who may respond to EGFR-TKIs and BRAF-targeted agents.
