Epidemiology
The ALDH2 gene, encoding acetaldehyde dehydrogenase 2, is located on chromosome 12. This enzyme possesses esterase, dehydrogenase, and reductase activities. Research has demonstrated that ALDH2 metabolizes nitroglycerin into nitric oxide, thereby promoting vasodilation and improving blood flow disturbances. However, ALDH2 gene polymorphisms are prevalent, especially in East Asian populations. The wild-type genotype ALDH2*1/*1 (GG) exhibits strong metabolic capacity. The heterozygous variant retains only 6% of the wild-type enzyme activity, while the homozygous mutant variant has virtually no enzyme activity, leading to extremely weak metabolism that cannot achieve the desired effect and may cause harm.
Work Flow
For genomic DNA extraction from EDTA-anticoagulated blood, a blood genomic DNA extraction kit is recommended.
A general DNA/RNA kit suitable for use with an automatic nucleic acid extractor may also be employed. The extraction must be performed strictly according to the manufacturer's instructions. The recommended elution volume is 100 µL.
