Epidemiology
The Zika virus is a single-stranded, positive-sense RNA virus of the Flaviviridae family, measuring 40–70 nm in diameter. It is enveloped, with a genome of 10,794 nucleotides encoding 3,419 amino acids. Genotypically, it comprises African and Asian lineages. Zika virus infection is a self-limiting acute disease, primarily spread via bites from Aedes aegypti mosquitoes. Symptoms typically include fever, rash, joint pain, or conjunctivitis, and fatalities are rare. According to the World Health Organization, neonatal microcephaly and Guillain-Barré syndrome may be linked to Zika virus infection.
Work Flow
Option 1: Use a viral RNA mini kit (nucleic acid extraction/purification reagent) and follow the manufacturer’s protocol, with a recommended input volume of 140 µL and an elution volume of 60 µL.
Option 2: Use a viral DNA/RNA kit together with an automated nucleic acid extraction system. Process according to the kit instructions, using a sample volume of 200 µL and an elution volume of 80 µL.

| Specificity | Use the kit to detect the serum samples with Zika virus negative, and the results are negative. Interference test results show that when the concentration of bilirubin in serum is not more than 168.2μmol/ml, the hemoglobin concentration produced by hemolysis is not more than 130g/L, the blood lipid concentration is not more than 65mmol/ml, the total IgG concentration in serum is not more than 5mg/mL, there is no effect on the dengue virus, Zika virus or chikungunya virus detection. Hepatitis A virus, Hepatitis B virus, Hepatitis C virus, Herpes virus, Eastern equine encephalitis virus, Hantavirus, Bunya virus, West Nile virus and human genomic serum samples are selected for the cross-reactivity test, and the results show that there is no cross reaction between this kit and the pathogens mentioned above. |
