Epidemiology
Ebola virus is a member of the Filoviridae family, characterized as an unsegmented, negative-sense, single-stranded RNA virus. Virions are elongated filaments, typically measuring about 1000 nm in length and 100 nm in diameter. The genome is roughly 18.9 kb in size and encodes seven structural proteins plus one non-structural protein. Five subtypes are recognized: Zaire, Sudan, Bundibugyo, Taï Forest, and Reston. The Zaire and Sudan subtypes are particularly lethal, having caused numerous human fatalities. Infection leads to Ebola hemorrhagic fever (EHF), an acute hemorrhagic illness transmitted primarily through contact with bodily fluids, secretions, or excretions from infected individuals or animals. Clinical features include high fever, hemorrhagic manifestations, and multi-organ failure. The case fatality rate ranges from 50% to 90%.
Work Flow
Option 1.
Utilize a viral RNA extraction kit or a nucleic acid purification reagent. Follow the manufacturer's instructions carefully. The recommended sample input volume is 140 µL, and the elution volume is 60 µL.
Option 2.
Use a viral DNA/RNA extraction kit paired with an automated nucleic acid extraction system. Adhere to the provided protocol. The sample volume should be 200 µL, and the elution volume is 80 µL.
Option 3.
Apply a nucleic acid extraction reagent and a high‑throughput automated sample preparation system. Extract as directed, with a sample volume of 160 µL and an elution volume of 60 µL.
